AnnexinII在胃癌组织中的表达及其在胃癌转移的作用

• 【摘要】探讨AnnexinII在胃癌组织中的表达及其在胃癌转移中的作用。方法：采用免疫组织化学法检测AnnexinII在51例胃癌组织和24例癌旁组织中的表达，并统计AnnexinII阳性表达与胃癌患者临床特征的关系。用AnnexinII特异性siRNA在胃癌HGC-27细胞抑制AnnexinII的表达，应用黏附实验、侵袭实验检测抑制AnnexinII表达后对胃癌细胞的黏附、侵袭能力的影响。结果：（1）AnnexinII蛋白在胃癌组织中阳性率为82.4%，在癌旁组织中阳性率37.5%,两组间差异具有显著性(P＜0.01)；并且AnnexinII阳性表达与患者的性别与年龄无关（P>0.05），而AnnexinII蛋白表达与肿瘤大小、组织分化程度、胃癌浸润深度、TNM分期、淋巴结转移等临床特征显著相关，差异具有统计学意义（P<0.05）。（2）在胃癌HGC-27细胞瞬时转染AnnexinII siRNA 24小时后， 细胞中AnnexinII 的蛋白和mRNA水平均显著下降，差异有统计学意义（p<0.05）。并且抑制AnnexinII 表达后，胃癌HGC-27细胞的黏附、侵袭能力显著下降，差异有统计学意义（p<0.05）。结论：AnnexinII在胃癌组织中高表达，AnnexinII蛋白表达与肿瘤大小、组织分化程度、胃癌浸润深度、TNM分期、淋巴结转移等临床特征显著相关。并且AnnexinII高表达可促进胃癌转移。因此，AnnexinII 可以作为抑制胃癌转移的潜在靶点，开发靶向 AnnexinII 的药物可能成为治疗胃癌转移的新方法。
• To investigate the expression of AnnexinII and their clinical significance in gastric cancer. Methods Expression of AnnexinII in 51 cases of gastric cancer and 24 normal gastric tissues was detected by immunohistochemistry staining. Then the correlation between the expression of AnnexinII in gastric cancer and the clinical significance were tested.RNA interference was used to downregulate the expressions of AnnexinII in human gastric cancera HGC-27 cells. Cell adhere test was used in AnnexinII siRNA transfected human HGC-27 gastric cancera cells to test cells adhesion potential. Cell invasion test was used in AnnexinII siRNA transfected human HGC-27 gastric cancera cells to test cells invasion potential. Results The positive expression rate of AnnexinII in gastric cancer was 82.4%, significantly higher than that in the normal gastric tissues( 37.5%, P<0.01). The positive expression AnnexinII has nothing to do with gender and age in gastric cancer (p>0.05). However, The positive expression of AnnexinII is correlated with tumor size, histodifferentiation, invasion,TNM classification, lymph nodes metastasisin in gastric cancer (p<0.05). Transfection of human gastric cancer cells with siRNA targetingAnnexinII, led to significant decreases in AnnexinII compared to cells transfected with the negative control siRNA. In AnnexinII siRNA transfected human HGC-27 gastric cancera cells, the numbers of attached cells, invaded cells were significantly decreased compared to cells transfected with the negative control siRNA..Conclusion AnnexinII is over-expressed in gastric cancer tissues and the positive expression of AnnexinII is correlated with the clinical significance of gastric cancer and .AnnexinII may play important roles in the metastasis of human gastric cancera cells.AnnexinII may enhance metabasis potential of human gastric cancera cells and may be a promoter of carcinogenesis. Thus, drug targetsAnnexinIImay present a new method for preventing gastric cancer metastasis in gastric cancer treatments.
