DJ-1在胰腺癌中的表达及临床意义

  • 【摘要】目的 探究DJ-1基因在胰腺癌中的表达以及和胰腺癌预后等的相关性。 方法 选取我院2012年12月-2015年3月肿瘤科住院部收治的术后病理确认为胰腺癌的患者64例为实验组,同时选取我院健康门诊体检者58例作为对照组,ELISA法检测DJ-1蛋白和CA19-9在所有入组者血清中的水平。统计分析不同TNM分期的胰腺癌患者血清中DJ-1的水平,并计算DJ-1水平与CA19-9水平的相关性。为探究不同水平的DJ-1和胰腺癌病程的关系,我们依据DJ-1水平对胰腺癌患者进行亚分组,对比不同DJ-1水平组胰腺癌病理资料。对术后的胰腺癌患者进行定期随访,进一步探究不同DJ-1水平和患者生存率的关系。 结果 ELISA结果水平发现64例胰腺癌患者血清中DJ-1蛋白水平显著高于58例正常人血清中DJ-1蛋白水平,差异具有明显的统计学意义(P<0.05)。分别统计不同胰腺癌亚分组中(T1 期,T2 期,T3 期,T4 期)血清中的DJ-1水平,结果显示四组DJ-1具有非常显著的差异,随着TNM分期的增加,DJ-1的水平显著升高。同时相关性分析发现血清DJ-1水平与CA19-9具有显著的正相关,回归系数r=0.78, 且高DJ-1组胰腺癌患者较低DJ-1组患者存在较高比例的肝脏转移和淋巴结转移,肿瘤大小以及TNM分期较低DJ-1组患者高。生存率分析结果显示高DJ-1组胰腺癌患者的生存率明显低于低DJ-1组胰腺癌患者,两组之间在术后14周后生存率之间的差异明显增加,高DJ-1组的生存率迅速降低,两组之间具有显著的统计学意义。 结论 DJ-1在胰腺癌患者血清中高表达,高表达DJ-1的胰腺癌患者癌症分期较高,生存率明显降低,这提示我们DJ-1水平可能为胰腺癌患者疾病预后的生物标志物之一。
  • Objective To explore the expression of DJ-1 in pancreatic cancer and its relation with pancreatic cancer prognosis. Methods Select 64 cases of pancreatic cancer in-patients in our hospital general surgery department during December 2012-March 2015 as experimental group, and at the same time choose health outpatient 58 cases as a control, ELISA method to detect serum CA19-9 and DJ-1 levels in two group. Statistical analyze serum DJ - 1 level in different TNM staging of pancreatic cancer patients, and explore different levels of DJ - 1 and the relationship between pancreatic cancer progression, calculate the correlation between DJ - 1 and CA19-9. To DJ - 1 level and level the correlation of CA19-9. We also compared pancreatic cancer pathology data according to the DJ - 1 level in patients with pancreatic cancer. Conduct regular follow-up in patients with postoperative pancreatic cancer to explore the survival of patients with different DJ-1 levels. Results ELISA found that DJ - 1 in 64 cases of patient serum with pancreatic cancer were significantly higher than that in 58 cases of normal human serum. DJ - 1 has obvious statistical significance difference (P < 0.05). Serum DJ-1 level in different pancreatic cancer group (T1, T2, T3, T4) has a very significant difference, significantly increased with the increase of TNM staging. At the same time correlation analysis found that serum CA19-9 and DJ-1 has significant positive correlation, regression coefficient r=0.78. pancreatic cancer patients with a lhigh DJ-1 level had a higher proportion of liver metastasis and lymphnode metastasis, tumor size, TNM staging was also high. Survival analysis showed that pancreatic cancer patients with high DJ-1 survival rate were significantly lower than that in pancreatic cancer patients with low DJ-1, quickly reduced survival rate in high DJ-1 group after 14 weeks, with significant statistical significance between two groups. Conclusions The serum level of DJ-1 was high in pancreatic cancer patients with a significantly lower survival rate, which suggested that DJ - 1 may be one of pancreatic cancer prognosis biomarkers.
