ALA-PDT对食管癌Eca-109凋亡相关基因表达的影响

- 【摘要】目的 凋亡相关基因Bak-1、FADD在食管癌Eca109细胞ALA-PDT的作用及初步探讨其在食管癌ALA-PDT凋亡通路的作用。方法 实验分为对照组和ALA-PDT组,治疗组细胞给予ALA(终浓度为0.25mM)孵育6小时后在630 nm Diomed 光动力治疗系统下进行光照(功率200 mW/cm2, 照光150 s,光剂量30J/cm2)。光照后0.5 h,3 h和24 h收集细胞,提取细胞RNA,采用Realtime PCR法检测Bak-1、FADD基因表达的变化。结果 FADD在ALA-PDT作用于Eca-109细胞3 h和24 h后其表达均显著下调(P均<0.05),Bak-1作用后0.5 h,3 h和24 h其表达相对对照组均下调,仅在作用后3 h有统计学差异,其余与对照组相比其差异无统计学意义(P均>0.05)。结论 Fas通路可能为ALA-PDT诱导Eca-109细胞凋亡的通路之一,FADD可能为该凋亡通路的靶基因之一。
- Objective To investigate the role of apoptosis related gene Bak-1 and FADD in ALA mediated PDT(ALA-PDT) in esophageal cell line Eca-109 and explore its role in apoptosis pathway in PDT. Methods The experiment was divided into two groups: the control group and PDT group. The cells of PDT group were incubated with ALA to the final concentration of 0.25 mM for 6 h in dark, and irradiated with the light dose of 30 J/cm2 (200 mW/cm2, 150 s) under the 630 nm laser system. Total RNA were extracted from Eca-109 cells at 0.5 h and 3 h and 24 h after ALA-PDT. Then quantitative real-time polymerase chain reaction(QRT-PCR) assay was used to detect the gene expression changes of Bak-1 and FADD. Rerults The expression of FADD was significant down regulation at 3 h and 24 h psot-PDT in the Eca-109 cell, and Bak-1 were down-regulation, at 0.5 h and 3 h and 24 h after treatment, but the difference contrast to the control group was not significant except at 3 h(P>0.05). Conclusion Fas pathway may play the role in the apoptosis way and FADD may be the target gene in ALA-PDT in Eca-109.
