MMP-11在乳腺浸润性导管癌肿瘤细胞中的表达与预后的相关性研究

- 【摘要】目的:研究MMP-11在乳腺浸润性导管肿瘤细胞的表达情况,探及MMP-11的表达与乳腺癌临床病理学指标及乳腺癌预后的相关性。 方法:本研究收集了2005-2008年总共285例诊断为浸润性导管癌的患者。应用免疫组织化学染色评估MMP-11在其肿瘤细胞中的表达情况,并收集患者的相关临床病理资料及随访信息。随后,评价MMP-11在乳腺癌中的表达情况及与临床病理学特征的相关性,并分析MMP-11的表达与乳腺癌预后的关系。 结果:MMP-11在肿瘤细胞中的表达与远处转移的出现显著相关(P<0.01)。MMP-11在肿瘤细胞中的表达和其他临床病理指标之间无相关性(P均>0.05)。单因素分析显示MMP-11在肿瘤细胞中的表达、组织学分级、脉管瘤栓、腋窝淋巴结情况、肿瘤分期及Ki67的表达均与无病生存期(DFS)和总生存率(OS)有相关性(P均<0.05),肿瘤大小与DFS有相关性(P<0.05)。多因素分析显示MMP-11在肿瘤细胞中的表达为DFS的独立影响因素。 结论:MMP-11在肿瘤细胞中的表达与乳腺癌预后的关系密切,对于预测预后意义重大。
- Objective: To investigate the correlation between the Matrix metalloproteinase-11 expression by tumor cells and clinicopathological features, and to evaluate whether MMP-11 expression was a predictor marker for prognosis in invasive ductal carcinoma of the breast . Methods: In this study, 285 invasive ductal carcinoma (IDC) patient samples were collected from 2005 to 2008. MMP-11 expression was examined in tumor cells by immunohistochemical staining, with clinical data and follow-up information from relevant patients collected. Subsequently, correlation analysis between IDC MMP-11 expression and clinicopathological features was performed, in addition to evaluating the relationship between MMP-11 expression and prognosis. Results: Tumor MMP-11 expression was significantly correlated with the presence of distant breast cancer metastases (P < 0.01). No significant correlation was found between tumor MMP-11 expression and other clinicopathological parameters (P > 0.05). Univariate analyses showed that tumor MMP-11 expression, tumor histological grade, vascular invasion, axillary lymph node metastasis, tumor stage and Ki67 expression were significantly associated with patient disease-free survival (DFS) and overall survival (OS) in IDC (P < 0.05), and tumor size were significantly associated with patient DFS (P < 0.05). Multivariate analysis showed that MMP-11 expression in tumor cells was independent factors affecting DFS, while it was not an independent factor affecting OS. Conclusion: MMP-11 expression in tumor cells was closely associated with IDC prognosis and significantly implicated as a potential predictive marker.
