PI3K/AKT信号通路相关蛋白在乳腺癌中的表达及其临床意义

- 【摘要】目的 研究PI3K/AKT信号传导通路中PTEN、PI3K、AKT蛋白在乳腺癌中的表达及其与临床特征、预后的相关性。方法 用免疫组化SP法检测45例乳腺癌和癌旁正常组织PTEN、PI3K、AKT表达。结果 PTEN、PI3K、AKT在乳腺癌中的表达率分别为22.2%、73.3%、75.5%,PTEN在乳腺癌中表达低于癌旁正常乳腺组织,而PI3K、AKT在乳腺癌中表达均明显高于癌旁正常乳腺组织( P<0.05);在乳腺癌组织中PTEN、PI3K、AKT的表达均与年龄无相关性( P>0.05);PI3K、AKT在乳腺癌中的表达和组织学分级、临床分期、腋窝淋巴结转移呈正相关( P<0.05),PTEN的表达和组织学分级呈负相关( P<0.05);PI3K、AKT的表达在有淋巴结转移时表达率较高,与无淋巴结转移组相比有统计学意义 ( P<0.05),PTEN的表达和淋巴结转移呈负相关( P<0.05),PTEN、PI3K、AKT的表达与临床分期、淋巴结转移有相关性( P<0.05)。结论 PI3K、AKT的过表达和PTEN的低表达和乳腺癌的形成和进展有关,检测PTEN、PI3K、AKT的表达可用于判定乳腺癌的恶性程度,并可作为判断乳腺癌预后的指标。
- Objective To investigate the expressions of PTEN, PI3K, AKT in breast cancer and their relevance with the clinical characteristics and prognosis of breast cancer. Methods The expression of PTEN, PI3K and AKT in 45 cases of breast cancer and non-neoplastic breast tissue was determined using SP method of immunohistochemeistry. Results The expression of PTEN, PI3K, AKT protein in breast cancer were 25.2%、70.3%、67.7% respectively, The expressions of PTEN protein in breast cancer lower than and in non-neoplastic breast tissue. While The expressions of PI3K and AKT protein in breast cancer higher than and in non-neoplastic breast tissue, ( P< 0.05).The expression of PTEN, PI3K and AKT protein did not correlate with age( P> 0.05).Positive correlation was found between the expression of PI3K, AKT and histological grade,clinical stage and axillary lymph node metastasis( P< 0.05),while negative correlation was found between the expression of PTEN and histological grade ( P< 0.05).The expression of PI3K and AKT was higher in lymph node metastasis than that in non lymph node metastasis, and the difference was siganificant ( P<0.05).While the expression of PTEN in lymph node metastasis was lower than that in non-lymph node metastasis. The expression of PTEN、PI3K and AKT in lymph node metastasis and clinical stage was correlation( P<0.05). Conclusion Overexpression of PI3K and AKT and low expression of PTEN may contribute to genesis and progression of breast cancer, detection of PTEN、PI3K and AKT may be useful in evaluation of the aggressiveness of breast cancer, and helpful to judge the prognosis of breast cancer.
