三氧化二砷对胶原诱导的血小板聚集的抑制作用机制研究

• 【摘要】目的 分析三氧化二砷是否影响胶原诱导的血小板聚集及其作用机制。方法 使用胶原诱导血小板聚集。使用光比浊法检测三氧化二砷对胶原诱导的抑制作用。使用流式细胞仪检测血小板聚集过程中表面GPIIb/IIIa变化。使用免疫分析试剂盒检测血小板聚集过程释放TXB2浓度。结果 胶原诱导的血小板聚集过程中，三氧化二砷明显抑制血小板TXA2的释放。三氧化二砷并未影响血小板聚集过程中血小板表面GPIIb/IIIa变化。结论 三氧化二砷通过抑制血小板TXA2激活路径抑制胶原诱导的血小板聚集。
• Objective To analyze whether arsenic trioxide has influence on collagen-induced platelet aggregation and its mechanisms. Methods Collagen was used to induce platelet aggregation. The inhibition of arsenic trioxide on platelet aggregation were measured by light aggregometry. The abundance of GPIIb/IIIa on the surface of the platelets during aggregation was measured by flow cytometry. The concentration of TXB2 was measured by an enzyme immunoassay kit. Results Arsenic trioxide inhibited TXA2 release from platelet during collagen-induced platelet aggregation. Arsenic trioxide did not affect GPIIb/IIIa on the surface of the platelets in the course of platelet aggregation. Conclusion TXA2 activation pathway of platelet is inhibited by arsenic trioxide in collagen-induced platelet aggregation.
